She saw which way they came from in the vehicle, Glen mentioned the prison and that it was a straight-shot, so she just followed the road and the signs. Simple as that.

 

No you guys are wrong... she used voodoo magic to find where the prison was.

 

She overheard Glenn and Maggie calling the prison home and how it was a straight show down the road from where they were at. Maggie also said you can hear the walkers banging on the fences from the woods.

I hadn't noticed that, so good spot, but I think she also has a map/maps in her kit-bag?
She had one in the first episode when she talked about getting to the coast. I'm not familiar with US road maps, so I'm only assuming tat it would be marked.
